This post is a bit dated but I found it only recently and thought it was useful enough to pass along. It seems elevator hacks have made the rounds on various blogs, thedamnblog, collegehumor.com, engadget, digg, etc.
Anyway, to take the express ride to your destination (on Otis and supposedly several other brand elevators - which presumably use the same or similar software) press your floor number and "close door" at the same time. This will bypass all the other requests and take you to your destination (skip floors waiting).
Also it has been rumored that if you press 3 floor buttons simultaneously it will clear all the previous selections. Go ahead, light up every floor and then press 3 floor buttons to clear it.
Note - Saw this in the comments on endadget:
i work for otis elevator and can honestly say this will not work on most elevators, its in the software but is only turned on if the customer wants it which they usually dont. there is something called independent service which u have to hold the door close button to close the door then push the floor and go non stop which is probably what this person who wrote the article witnessed, but you have to have a key to turn it on :) which i have all of them. i go express every trip.
